【摘要】:Thesetestmethodsareusedtodemonstratewhetheranadhesivepreparationissufficientlyprotectedwithbiocidetoresistattackbybacteria,yeast,andfungiduringitsstoragelife.Theyarepatternedaftermethodsusedbybiologicallaboratoriesservingtheadhesiveindustry.Thesetestmethodsmayalsobeusedtodeterminetheefficacyofdifferentbiocidesystemsagainstspecificmicroorganisms.Thesetestmethodsareespeciallyusefulwhentestedagainstwild-typemicroorganismswhichhavebeenisolatedfromcontaminatedadhesivesasanaidindeterminingtheamountandtypeofbiocidenecessarytokillorinhibitthegrowthofthecontaminants.Ifanisolatedmicroorganismnotgenerallyusedasachallengeorganism,ischosenastheinoculum,itisimportanttoidentifytheorganismanddetermineonwhichmediumandunderwhatconditionsitwillgrow,inordertodemonstratetheefficacyofthebiocide.Theresultsobtainedwhenusingtheproceduresgiveninthesemethodsapplyonlytothespecieswhichareusedforthetesting.ThetestspecieslistedinSection9arefrequentlyusedbylaboratoriestotestforantimicrobialproperties,buttheyarenottheonlyoneswhichcouldbeused.Selectionofthespeciestouseforthesetestmethodsrequiresinformedjudgmentbythetestinglaboratoryorbythepartyrequestingthetests.Itisalsoimportantthatspecieswhichcommonlyattackadhesivesbeused.See9.4.Thepresenceofanactivebiocidecarriedoverfromtheadhesivespecimentotheagarcouldhaveaninhibitingeffectonthegrowthofmicroorganisms,resultinginnogrowthduringthespanofanormalincubationperiod,wheninfact,viablemicroorganismsarepresent,buttheirgrowthhasbeensloweddownorheldinstasis.TheuseofLetheenagarandbrothisrecommendedtoneutralizetheeffectofthiscarry-over.Note48212;Letheenagarmaybeusedforthestreakplates,orifanotheragarischosenfortesting,aLetheenagarplatecouldbestreakedasacontroltotestagainsttheneutralizingeffect.EvenmoreeffectivewouldbedilutingthechallengedadhesivespecimenwithLetheenbrothandrunningLetheenagarpourplates.SeeNote1andNote3.Extendingtheincubationperiodofnegativeplateswouldbeanothersafeguard.Toneutralizethiazoline-basedpreservatives,10to50ppmofsodiumthioglycolatecanbeaddedtothemedium.Thesetestmethodsaredependentuponthephysiologicalactionoflivingmicroorganismsunderareportedsetofconditions.Conclusionsabouttheresistanceofthetestadhesivetomicrobiologicalattackcanbedrawnbycomparingtheresultstosimultaneouslyruncontrolsofknownresistance.SeeX5.2forstatementsregardingtestrepeatability.1.1Thesetestmethodscoverthedeterminationoftheresistanceofliquidadhesivepreparationstomicrobialattackinthecontainerbychallengingadhesivespecimenswithculturesofbacteria,yeast,orfungi,andcheckingfortheirabilitytoreturntosterility.Thesetestmethodsreturnqualitativeresults.1.2ThevaluesstatedinSIunitsaretoberegardedasthestandard.Thevaluesinparenthesesareforinformationonly.1.3Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.Thesetestmethodsaredesignedtobeusedbypersonstrainedincorrectmicrobiologicaltechnique.SpecificprecautionarystatementsaregiveninSection8.
